摘要
目的:研究碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)对体外培养人牙髓细胞增殖活性的影响。方法:以改良组织块培养法体外获得人牙髓细胞,以细胞增殖检测试剂盒(cell counting kit-8,CCK-8)研究bFGF在不同浓度(1.0、5.0、10.0和50.0ng/mL)、不同时间(1、2、4和7d)作用下,牙髓细胞增殖活性的变化。结果:bFGF在1.0~50.0ng/mL浓度下可促进体外培养人牙髓细胞的增殖(P<0.05),bFGF促进牙髓细胞增殖的最低显效浓度为1.0ng/mL,最佳显效浓度为10.0ng/mL。从第2天起,bFGF可诱导体外培养人牙髓细胞增殖(P<0.05),bFGF诱导牙髓细胞增殖在第4天即可达到高峰(P<0.01)。结论:bFGF可诱导体外培养人牙髓细胞增殖,该诱导作用具有剂量依赖性和时间效应。
Objective To investigate the effect of basic fibroblast growth factor (bFGF) on the proliferation of in vitro cultured human dental pulp cells. Methods Human dental pulp cells were isolated from dental pulp tissue explants. The effects of bFGF treatment at concentrations of 1.0 ~ 50.0 ng/mL for 1 ~ 7 d on the proliferation of dental pulp cells were assayed with a cell counting kit-8(CCK-8). Results bFGF at concentrations of 1.0 ~ 50.0 ng/mL markedly promoted the proliferation of dental pulp cells(P 〈0.05). The promoted effect of bFGF on the proliferation of human dental pulp cells maximized at a concentration of 10.0 ng/mL and 4 days after treatment (P 〈0.01). Conclusion bFGF can promote the proliferation of human dental pulp cells in a dose-and time-dependent manner.
