摘要 目的:探讨维生素C(VitC)对人肝癌细胞SMMC-7721增殖的影响和作用机制,从而为肝癌治疗的临床用药提供参考。方法:MTT法检测VitC对人肝癌细胞SMMC-7721增殖的影响,利用Hoschest核染和荧光显微镜观察肝癌细胞SMMC-7721的形态学变化,同时通过流式细胞仪测定VitC对肝癌细胞SMMC-7721各组周期变化与凋亡分析。结果:VitC对人肝癌细胞SMMC-7721的增殖有抑制作用,且有浓度和时间依赖性;在荧光显微镜下观察细胞形态,分别出现典型的凋亡细胞形态特征;流式方法测定的结果显示VitC对阻滞肝癌细胞SMMC-7721于S期,抑制其生长并诱导凋亡。结论:VitC对人体肝癌细胞SMMC-7721生长有抑制作用,使其细胞周期阻滞于S期并诱导其凋亡。 Objective To study the effects of vitamin C (VitC) on the proliferation and apoptosis of hepatocellular carcinoma cells SMMC-7721. Methods MTT assay was used to detect the proliferation,hoechst staining and fluorescence microscopy were used to examine the morphologic changes,and flow cytometry was used to measured the apoptosis status of hepatocellular carcinoma cells SMMC-7721. Results The proliferation of hepatocellular carcinoma cell SMMC-7721 was inhibited concentration-and time-dependently by VitC treatment. After treated by VitC,typical morphological characteristics of apoptosis were observed under fluorescence microscopy in cell morphology,SMMC-7721 liver cancer cells were stayed in S phase,and apoptosis was induced. Conclusion VitC can inhibit the proliferation of hepatocellular carcinoma cell SMMC-7721,induce the cell cycle arrest in S phase and apoptosis.
