摘要
通过考察抽提野生余甘子的缓冲液pH、抽提体积和抽提温度进行优化,探讨余甘子SOD的最佳提取工艺.结果表明:抽提缓冲液的pH7.8,抽提体积为2倍,抽提温度为4℃时,得到余甘子SOD的提取的粗酶活性为198.47 U/mL(。NH4)2SO4饱和度为80%时,余甘子SOD能够达到相对最大酶活性。
The optimal technology of extracting SOD from wild Fructus Phyllanthi was studied by optimizing pH value,bulk multiple and extraction times of phosphate bufer and solution bufer.SOD was calculated by the method of Pyrogallic acid auto-oxidation.The results showed that the activity of SOD was highest when applying the condition of pH 7.8,extraction time 2 and extraction temperature 4 ℃.The optimal saturation of ammonium-sulfate for the highest activity of SOD is 80%.
