摘要
目的观察糖皮质激素受体阻断剂(RU38486)对冷束缚应激状态(cold restrain stress,CRS)下大鼠肠屏障功能改变的影响;探讨糖皮质激素受体对CRS状态下大鼠肠屏障功能的影响机制。方法将60只成年雄性SD大鼠,随机分为正常对照组、应激组(S组)和糖皮质激素受体阻断剂组(R组)。S组和R组按应激后检测时间又分为S2h、S4h、S8h、S16h、S24h组和R2h、R4h、R8h、R16h、R24h组。应激动物模型参照Brodie[1]的方法改良制作。动态观察各组大鼠肠黏膜屏障功能的改变情况。结果应激组各时段大鼠血清D-乳酸浓度、血浆内毒素浓度及肠黏膜损伤指数均高于正常组,而糖皮质受体阻断后血清D-乳酸浓度、血浆内毒素浓度及肠黏膜损伤指数均较应激组相应各时段明显增高。结论糖皮质激素受体阻断剂可加重应激引起的肠黏膜上皮的损伤,导致肠黏膜屏障通透性增高,造成肠屏障功能的损害。
Objective To observe the effect of the glucocorticoid receptor blockage on the changes of intestinal barrier function in rats with cold restrin stres(CRS) and discuss its possible machanisms.Methods The SD rats were divided into three groups randomly:normal group,stress group and glucocorticoid recrptor blockage group.The stress model was induced in terms of the model by brodie.Blood samples were taken for examination of the endotoxin.D-lactate at various intervals after stress.Biopsy specimens were processed for routine histological examination and electron microscopic examination.Results The experimental results indicated that the blood plasma endotoxin level,the serum D-lactate level and the mucosa injury degree in both strees(S) group and glucocorticoid receptor blockage(R) group increased after stress.Compared the factors all corresponding intrevals of group S,all of the date of group R was much higher than group S(P0.05).Conclusion The glucocorticoid receptor blockage aggravates the injury of intestinal mucosa,then leads to impairment of intestinal barrier function.
