摘要
骨关节炎(osteoarthritis,OA)、大骨节病(Kaschin-Beck disease,KBD)等退行性骨关节病,均表现为软骨细胞外基质的过度降解和丢失。早期研究发现,O A和K B D病人软骨和关节滑液中糖醛酸含量增加,病人体内硫酸化程度降低。近年,糖生物学已成为生命科学的新前沿,研究提示糖复合物糖链修饰的改变与很多疾病的发生相伴随。蛋白聚糖(proteoglycan,PG)是一种在人和动物组织中分布广泛且重要的糖复合物,本文拟就聚集蛋白聚糖(aggrecan,A G)的结构及其代谢中硫酸化修饰与骨关节病研究进行综述,以期为骨关节病的深入研究和治疗提供新思路。
Osteoarthritis(OA) and Kaschin-Beck disease(KBD) are all retrogressive osteoarthrosis,manifested as excessive degradation and loss of extracellular matrix of articular cartilage.Early study found that increased uronic acid content existed in cartilage and synovial fluid of patients with OA and KBD,and the sulfation degree was found lower in these patients.In recent years,glycobiology has become the new frontier of life science.Some studies show that changes in glycoconjugates chains accompanied by occurrence of many disease.Proteoglycan is a widespread and important glycoconjugate.This review summarized the study of structure and sulfated modification in metabolism of aggrecan and osteoarthrosis with the objective to provide new ideas for further study and therapy of osteoarthrosis.
