摘要
对多孔壳层Halo-C_(18)色谱柱、Zorbax Eclipse XDB-C_(18)色谱柱、Zorbax SB-C_(18)色谱柱、ACQUIT UPLC BEH-C_(18)色谱柱以及多孔壳层Halo-C_(18)与Zorbax SB-C_(18)的串联色谱柱分离分析头孢菌素类药物进行了比较研究。结果表明:多孔壳层Halo-C_(18)色谱柱的分离效果明显优于Zorbax Eclipse XDB-C_(18)色谱柱和Zorbax SB-C_(18)色谱柱。串联柱的柱效、分离度几乎达到ACQUITUPLC BEH-C_(18)色谱柱的水平,而其柱压却只有ACQUIT UPLC BEH-C_(18)色谱柱的一半。多孔壳层Halo-C_(18)与Zorbax SB-C_(18)串联色谱柱在一定程度上可以取代超高压色谱柱在常规液相色谱仪上实现高通量快速分离。
Five C_(18)-bonded silica chromatographic columns,i.e.,the①Halo-C_(18) column,②Zorbax Eclipse XDB-C_(18) Column,③Zorbax SB-C_(18) Column,④ACQUIT UPLC BEH-C_(18) column and⑤coupled column of Halo-C_(18) and Zorbax SB-C_(18) column were used separately in the separation of 6 cephalosporins with gradient elution and their separation efficiency were studied comparatively.It was found that the separation efficiency of the column①was significantly higher than that of the columns②and③;and that the column efficiency and resolution of the coupled column⑤attained to nearly the same levels as those of the column④,but the column pressure of column⑤was only half of the column④.It was obvious that the coupled column⑤could be used instead of the column④,i.e.,the ACQUIT UPLC BEH-C_(18) column,to a certain extent in the rapid HPLC analysis of cephalosporins.
