摘要
本文研究了六种大孔吸附树脂HZ801、HZ806、HZ816、HZ818、HZ830和AB-8对叶黄素酯的吸附和解吸性能,结果表明HZ816较其他树脂表现出更好的吸附和解吸能力,并对该树脂的静态和动态吸附解吸条件进行了研究,确定了树脂纯化叶黄素酯的最佳工艺参数。最佳工艺条件为:在25℃下,以20%正己烷-乙醇为吸附溶剂,上样液浓度在1.432-1.891mg/mL范围内,吸附流速2BV/h,上样量7BV;洗脱剂为乙醇和50%正己烷-乙醇溶液,洗脱流速为3BV/h。经树脂纯化后,叶黄素酯的纯度由63.3%提高到91.7%,回收率为75.9%。本文还对HZ816树脂的重复利用性做了研究,对同一树脂柱进行10个周期的重复实验,结果表明树脂的吸附量和解吸率较初始时并没有显著下降,亦无破碎现象,树脂稳定性良好,可在生产中多次重复利用。
In present study,the performance and separation characteristics of six macroporous resins for purification of lutein ester from Tagetes erecta have been evaluated.These resins are HZ801,HZ806,HZ816,HZ818,HZ830 and AB-8.The results showed that HZ816 offered higher adsorption and desorption capacities.The static and dynamic adsorption and desorption was studied and optimization of the purification process of lutein ester from Tagetes erecta was performed.The best conditions were: the volume,concentration,adsorption sovlent and flow rate of loading sample were 7 bed volumes(BV),1.432-1.891mg/mL,20%hexane-ethanol and 2BV/h,respectively,at 25℃;desorption was performed successively with ethanol and 50%hexane-ethanol.After once purification,the lutein ester content in the product was increased from 63.3% to 91.7% with a recovery of 75.9%.The resin reusable study showed that after 10 times purification on the same resin column,the adsorption capacity and desorption rate did not decrease significantly.Resin didn't break as well.Therefore,it can be reused in the industry.
