摘要
目的:研究5'-核苷酸酶(5'-NT)在不同类型白血病、骨髓增生异常综合征(MDS)、非白血病患者和正常人中的分布特点。方法:分离100例不同白血病、3例MDS患者、9例非白血病患者和6例正常人骨髓有核细胞,以5'-单磷酸胞嘧啶核苷(5'-CMP)为底物进行细胞组织化学反应,电镜半定量分析5'-NT阳性细胞率和阳性指数。结果:5'-NT阳性反应产物主要位于有核细胞胞膜。急性髓系白血病(AML)M1、M2a、t(8;21)、t(15;17)各组细胞阳性率和阳性指数依次降低,阳性率组间差异有统计学意义(P<0.01),阳性指数差异无统计学意义;M1、M2a、t(8;21)、t(15;17)和M5患者5'-NT阳性细胞率和阳性指数均显著高于MDS、非白血病患者和正常人,差异有统计学意义(P<0.01);急性B淋巴细胞白血病(B-ALL)患者5'-NT阳性细胞率和阳性指数高于慢性淋巴细胞白血病(CLL),差异有统计学意义(P<0.05);CLL、MDS、非白血病患者和正常人的5'-NT阳性细胞率和阳性指数低,组间及两两比较差异无统计学意义。结论:不同白血病患者胞膜5'-NT表达与白血病细胞分化程度负相关。
Objective:To study distribution of 5'nucleotides(5'-NT) in patients with various leukemia,MDS,non-leukemic diseases,and healthy adults.Method:The nucleated cells were isolated from bone marrow of 100 patients with various leukemia subtypes,3 of MDS,9 of non-leukemic patients and 6 healthy donors.The reactivity of 5'-NT was demonstrated by cytochemistry of 5'-cytidine monophosphate(5'-CMP) .The positive cell ratio and their index were measured with electron microscope on the base of ultrastructural observation semiquantitatively. Result:The positive product was located on plasma membrane.Positive ratios and indexes of M1,M2a,t(8;21) and t(15;17) were decreased in orderly.Their positive ratios were different statistically(P0.01) ,but positive indexes not.The positive ratios and indexes of all AML subtypes were higher than MDS,non-leukemia and healthy donors statistically(P0.05) .The positive ratio and index of B-ALL were higher than CLL statistically(P0.05) .The positive ratios and indexes of CLL,MDS,non-leukemic patients and healthy donors were lower and no significant difference between them.Conclusion:The results suggested 5'-NT expression was an important index of blood cells,correlated with leukemic cell differentiation negatively.
