脂质体槲皮素对糖尿病大鼠视网膜糖基化终产物生成及受体表达的影响 被引量:5

Effect of liposomal quercertin on advanced glycation end products and receptor for advanced glycation end products in retina of diabetic rats

摘要 目的观察脂质体槲皮素(LQ)对糖尿病(DM)大鼠视网膜糖基化终产物(AGEs)生成及受体(RAGE mRNA)表达的影响。方法采用蒸发-高压均质法制备LQ悬浊液,链脲佐菌素(STZ)法诱导DM大鼠模型,并随机分为DM模型组、LQ低、中、高剂量及氨基胍灌胃组;连续给药12周后,免疫荧光法、RT-PCR方法分别检测DM大鼠视网膜中AGEs沉积情况及RAGE mRNA的表达。结果与DM模型组结果(AGEs:0.922±0.005;RAGE mRNA:1.192±0.098)比较,LQ各组及氨基胍组视网膜中AGEs沉积减少,RAGE mRNA表达也降低(P<0.05),以LQ中剂量组(AGEs:0.734±0.007;RAGE mRNA:0.724±0.028)降低最为明显(P<0.01)。结论 LQ可通过有效减少DM大鼠视网膜中AGEs的沉积,下调RAGE mRNA的表达。 Objective To investigate the effect of liposomal quercertin on advanced glycation end products(AGEs) and receptor for advanced glycation end products (RAGE) in retina of diabetic rats. Methods Liposomal quercetin was prepared by thin-film evaporator and high pressure homogenization method. Experimental diabetic rats induced by streptozotocin (STZ) were randomly divided into the following groups:diabetes mellitus (DM) control group, liposomal quercetin low, middle and high doses groups, aminoguanidine control group. After 12 weeks, the expression of AGEs and RAGE mRNA in retina were detected through immunofluorohistochemistry and reverse transcription polymerase chain reaction (RT- PCR). Results The level of AGEs in liposomal quercetin groups and aminoguanidine group was lower than that of diabetes mellitus control group (AGEs:537.29 ± 103.21 ). The level of RAGE mRNA (1. 192 ± 0. 098) in liposomal quercetin groups and aminoguanidine group was lower than that of diabetes mellitus control group, and the liposomal quercetin middle dose group (AGEs: 478.75 ± 90.34; RAGE mRNA: 0.724 ± 0. 028 ) decreased significantly ( P 〈 0. 01 ). Conclusions Liposomal quercetin can intervene the development of retinal microangiopathy in early period by reducing the level of AGEs and RAGE mRNA in retina diabetic rats effectively.
