摘要
目的探讨野菊花中黄酮类成分抗氧化有效部位最佳提取工艺,并考察野菊花不同提取部位的抗氧化活性。方法采用预试验,分别考察了不同提取方法,不同洗脱部位对结果的影响。选用L9(34)正交试验对黄酮有效部位的提取工艺进行研究。并采用DPPH法,比较野菊花不同提取工艺的抗氧化活性。结果预试验中各因素均对结果有影响,最好方法为超声提取,30%乙醇洗脱。正交试验中影响总黄酮得率的因素依次为:溶媒浓度,提取次数,固液比,提取时间。最佳工艺为:40%乙醇提取,1∶12的加液量,超声提取2次,每次40min。结论该实验确定的最佳提取工艺稳定性较好,简单可行。
Objective To investigate the best technological condition to extracting flavonoid from flos chrysanthemi indici,and to investigate the antioxidant activity of different extracted parts.Methods By using pre-experimental design,the effects of different chromatography,different extraction method and different smashed degree were obtained.We used L9(34) orthogonal experimental design to investigate the better condition of extraction.DPPH method used to compare different wild chrysanthemum extraction of the antioxidant activity.Results the factors of the pre-experiment have obvious effect.The best way for ultrasound extraction,macroporous resin chromatography,elution 30% ethanol.Orthogonal effects of flavonoids in the yield factors were: solvent concentration,extract the number of solid-liquid ratio,the extraction time.The best technology: 40%ethanol extraction,1∶12 amount of the increase,from 2 ultrasound,each for 40 minutes.Conclution The best technological condition to extracting total flavones from Chrysanthemi indici flos is stable,simple and feasible.
