三种神经细胞黏附分子在人脑星形细胞瘤中表达的差异

Expression difference of neural cell adhesion molecule in astrocytoma

摘要 目的了解三种神经细胞黏附分子(120kD、140kD、180kD)在不同级别人脑星形细胞瘤中表达的差异及相关性。方法针对三种神经细胞黏附分子分别设计引物,对54例标本进行了RT-PCR、免疫组织化学和病理分级,寻找三种分子在RNA和蛋白质水平上的表达规律。结果三种mRNA中仅140kD在星形细胞瘤中存在表达下调,但在不同病理分级的组间无统计学差异;神经细胞黏附分子主要分布在胞质和胞膜,随着肿瘤恶性程度的增加,神经细胞黏附分子的合成量亦明显下降。结论在三种mRNA中140kD的作用最为关键,神经细胞黏附分子的表达程度可以作为星形细胞瘤病理分级的一个重要指标。 Objective To find out NCAM expression difference in astrocytoma at mRNA and protein level. Methods Total RNA were extracted in 43 astrocytoma and 1 U251, after purifiy through three pair primers (NCAM-180, NCAM-140, NCAM-120) RT-PCR were used to found out their expression difference at RNA level. At the same time immuno-histochemical analysis were also taken to discover the expression difference at protein level. Results Among the three kinds of mRNA only NCAM-140' expression declined in astrocytoma(F = 4.72 ,P 〈 0.01 ). NCAM protein's expression was reduced with the rise of astocytoma pathological elassificationon. Conclusions NCAM-140 was the most important mRNA among the three isoform. NCAM protein can be used as a pathological classificationon index in astrocytoma.
