端粒重复序列结合因子1和2及端粒保护蛋白1基因在食管癌中的表达研究

Studies on the expressions of TRF1,TRF2 and POT1 in esophageal carcinoma

目的检测端粒保护蛋白TRF1、TRF2和POT1基因mRNA在食管癌组织中的表达。方法收集33例明确诊断的食管癌中心组织及其相应癌旁组织,提取总RNA并逆转录成cDNA,采用SBYRGreen适时荧光定量PCR方法检测TRF1、TRF2和POT1基因在食管癌中心组织和癌旁组织mRNA水平的表达。结果 TRF1mRNA在食管癌中心和癌旁表达所得CT值与β-actinmRNA所得CT值之差(即ΔCT值)分别为1.42±2.31和1.02±1.04,差异有统计学意义(P<0.05,n=33);POT1在两者中的表达分别为4.72±1.47和3.80±1.06,两者差异有统计学意义(P=0.001,n=33);TRF2为8.53±2.57和8.38±1.50,两者差异无统计学意义(P>0.05,n=33)。结论虽然TRF2mRNA在食管癌组织中和肿瘤旁表达没有差异,但TRF1和POT1mRNA在食管癌组织中表达异常降低,提示TRF1和POT1可能与食管癌的发生、发展有密切关系。

Objective To determine repeat binding factor 1 （ TRF1） ,factor 2 （ TRF2） and protection of telomeres1 （ POT1） mRNA expression in esophageal cancer. Methods The cancer center tissues and adjacent tissues of esophageal carcinoma were collected by surgery. The total RNA was extracted by Trizol and then reversed transcript into cDNA to preserve. SYBR green fluorescence quantitative PCR was used to detect the mRNA expressions of TRF1,TRF2 and POT1. Results The CT differential values （ ΔCT values） between TRF1 CT values and β-actin CT values were 1. 42 ± 2. 31 and 1. 02 ± 1. 04 in esophageal cancer center tissues and adjacent tissues respectively. The expressions of TRF1 mRNA in esophageal cancer center tissues are lower than that of adjacent tissues （ P 〈0. 05,n =33）. The ΔCT values of POT1 were 4. 72 ±1. 47 and 3. 80 ±1. 06 in both respectively. The expressions of POT1 mRNA in esophageal cancer center are lower than that of adjacent tissues （ P =0. 001,n =33）. The ΔCT values of TRF2 were 8. 53 ±2. 57 and 8. 38 ±1. 50 respectively,and there were no difference between them （ P 〉0. 05,n = 33）. Conclusions Although there was no significant difference between TRF2 mRNA in esophageal cancer tissues and its adjacent tissues,the abnormal lower of TRF1 and POT1 mRNA expression occurred in tissues of esophageal cancer,which suggests that TRF1 and POT1 are associated with the development of esophageal cancer.
