椰子溶磷脂酸酰基转移酶基因克隆及农杆菌介导甘蓝型油菜的遗传转化

摘要 月桂酸是在清洁用品和医药产品中有重要用途的中链脂肪酸。椰子中富含月桂酸特异的溶磷脂酸酰基转移酶基因,克隆该基因在油菜中表达若可获得含月桂酸的油菜新种质,将具有良好的应用价值。为获得脂肪酸成分改良的油菜种质资源,从椰子胚乳中克隆出该基因并构建35S驱动的植物表达载体,通过农杆菌介导法转化油菜子叶柄,在获得的约20株的转化苗中已鉴定有6株转化植株。 Laural acid is an important medium-chain fatty acid in cleaning supplies and pharmaceutical products.Coconut has abundant Lyso-phosphatidic acid acyltransferase(LPAAT) genes with expression of laural acid.New rape germplasm resources with Lyso-phosphatidic acid acyltransferase genes will have the good application value.The paper reports that Lyso-phosphatidic acid acyltransferase gene is cloned from coconut endosperm,the plant expression vector by 35S drive is built successfully,and 6 rape plants with Lyso-phosphatidic acid acyltransferase gene are selected from 20 genetic transformation rape plants by agrobacterium-mediated method.
