摘要
采用HPLC技术对盐藻中β-胡萝卜素顺反异构体进行分分析,探索适宜的分析条件.所用仪器为WatersHPLC分析系统,检测波长λ=436nm;灵敏度AuFs0.05;色谱柱为:250×4.6C18柱(Nucle-osi7μm);洗脱条件为:甲醇∶氯仿=60∶40(V/V),流速为1mL/min.所有操作过程均在避免高温、高湿度、强光照射、低pH值条件下进行.
Carotenoids have big family in which β carotene have mang different stereoisomers.Our experiment using high performance liquid chromatography to analyse the cis trans isomers of Dunaliella salina β carotene,in order to get the optimun analylical conditions.We use WATERS HPLC system to get suitable conditions:1.Chromatographic columns:C 18 (250×4.6,Nucleosi 7μm);Mobile phase:Methanol∶Trichloromethane=60∶40( V/V );Flow rate:1 mL/min;detection,λ=436nm;temperature at 0~10℃.2.Two kinds of cis isomers and trans isomers are found.3.All of the experiments are in the conditions avoiding high temperature,stong,light,low pH.
