雌激素与植物雌激素对卵巢切除大鼠乳腺和血管核因子-κB表达的影响 被引量:2

Effects of estrogen and phytoestrogens on the nuclear factor-κB expression in breast tissue and aorta of ovariectomized rats

摘要 目的探讨雌激素与植物雌激素对卵巢切除大鼠乳腺和血管核因子-κB(NF-κB)表达的影响。方法分别给卵巢切除大鼠皮下注射17β-雌二醇、17β-雌二醇合并黄体酮、黄体酮、染料木素、白藜芦醇或根皮素21 d,免疫组织化学法观察不同处理组大鼠乳腺和血管组织NF-κB的表达,以假手术大鼠作为对照。结果 NF-κB在乳腺中表达定位于乳腺导管上皮细胞的细胞质内;在血管壁中表达定位于内膜内皮细胞和中膜血管平滑肌细胞的细胞浆和(或)细胞核内。卵巢切除后表达降低,给予外源性的雌二醇、雌二醇合并黄体酮或黄体酮后表达增高;3种植物雌激素组表达无明显变化。结论雌、孕激素明显上调去卵巢大鼠乳腺、血管NF-κB的表达,植物雌激素染料木素、白藜芦醇和根皮素对其形态学和NF-κB的表达无明显影响。 Objective To explore the expression of nuclear factor-κB(NF-κB) in breast tissue and aorta of ovariectomized(Ovx) rats after treated with estrogen,phytoestrogens.Methods Ovx rats were subcutaneously given 17β-estradiol,17β-estradiol combined with progesterone, progesterone,or genistein,resveratrol and phloretin for 21 days.Taking sham operation rats as a comparison,the expression in breast tissue and aorta of NF-κB was examined by immunohistochemical technology.Results The expression of NF-κB in breast tissue located in cytoplasm of ductal epithelial cells.The expression of aorta located in cytoplasm(or) nucleus of vascular intima endothelial cells and of vascular smooth muscle cells.Every group had different expressions. The expression in group Ovx was sharply reduced compared with that of group sham; compared with the expression of group Ovx,the expressions of group Ovx+17β-estradiol,group Ovx+17β-estradiol+progesterone and group Ovx+progesterone were all increased significantly. But the groups of genistein,resveratrol and phloretin had no obvious change.Conclusion After having been treated with estrogen and progesteron,the expression of NF-κB is up-regulated. However,genistein,resveratrol,phloretin has no effect on the expression of NF-κB in breast tissue and aorta of Ovx rats.
