Triterpenoids from the Fruits of Forsythia suspensa

目的:对连翘(Forsythia suspensa (Thunb.) Vahl.)的化学成分进行研究。方法:利用反复硅胶和凝胶 LH-20, ODS柱层析进行分离和纯化, 通过理化性质和波谱数据分析鉴定化合物结构。结果:分离并鉴定出 11 个化合物, 分别为3β-acetoxyl-20S-dammarane-23-ene-20, 25-diol (1), dammar-24-ene-3β-acetate-20S-ol (2), 3β-acetoxyl-20S, 24R-dammarane-25-ene- 24-hydroperoxy-20-ol (3), β-amyrin acetate (4), betulinic acid (5), morolic acid (6), phillyrin (7), pinoersinol (8), phillygenin (9), 3β-acetyloleanolic acid (10), 3β-hydroxy-labda-8(17), 13E-dien-15-oic acid (11)。我们对化合物 1-3 进行了细胞毒试验, 均显示出较强的抑制胃癌细胞增殖的活性。结论:化合物 1,3, 6, 11 为首次从木樨科植物中分离得到, 其中化合物 2 显示了显著的细胞毒活性。

AIM： To investigate the chemical constituents from the fruits of Forsythia suspense （Thunb.） Vahl.. METHODS： Compounds were separated by column chromatography with silica gel, Sephadex LH-20 and ODS. Their structures were elucidated on the basis of physiochemical and spectral analyses. RESULTS： Eleven compounds were obtained from the ethyl acetate fraction, and their structures were elucidated as 3-acety-isofouquierol（1）, dammar-24-ene-3β-acetate-20S-ol （2）, 3β-acetoxyl-20S, 24R-dammarane-25-ene-24-hydroperoxy-20-ol （3）, β-amyrin acetate （4）, betulinic acid （5）, morolic acid （6）, phillyrin （7）, pinoersinol （8）, phillygenin （9）, 3β-acetyloleanolic acid （10）, and 3β-hydroxy-labda-8 （17）, 13E-dien-15-oic acid （11）. We further tested the cytotoxity of the three dammar triterpenoids. They all showed cytotoxity against gastric cancer cells. CONCLUSION： Compounds 1, 3, 6, 11 were isolated from the Forsythia genus for the first time. Compound 2 was the most potent proliferation inhibition compound.
