摘要
目的研究人脑胶质瘤中端粒酶活性与P53蛋白表达的情况,并探讨其与胶质瘤的相关关系及可能存在的临床意义。方法采用PCR-ELISA法和免疫组化ABC法,对72例人脑胶质瘤及16例正常脑组织标本中的端粒酶活性和P53蛋白分别进行检测。结果脑胶质瘤中端粒酶活性和P53蛋白的阳性表达率分别为61.11%(44/72)和51.39%(37/72),且各组阳性率随病理级别的升高而增加(P<0.01),而正常脑组织中两者表达均为阴性;P53蛋白表达阳性患者端粒酶活性表达的阳性率为83.78%(31/37);P53蛋白表达阴性患者端粒酶活性表达的阳性率为37.14%(13/35),两者之间存在相关关系(P<0.05)。结论 P53蛋白与端粒酶共同参与了脑胶质瘤的发生发展过程,可作为反映胶质瘤恶性程度的重要指标。
【Objective】 To study the expression of telomerase activity and P53 protein in human glioma and to investigate the correlations between their expression and clinical significance.【Methods】 Telomerase activity was detected by PCR-ELISA and immunohistochemistry ABC method was used to examine the P53 protein both in glioma tissue of 72 cases and brain tissue of 16 normal cases.【Results】 The positive rates of telomerase activity and P53 protein in brain glioma were 61.11%(44/72) and 51.39%(37/72) respectively,which were positively correlated with the pathological type(P 0.01),but no telomerase activity and P53 protein were detected in 16 normal brain tissues.Furthermore,the expression of P53 protein was significantly positively associated with telomerase activity(31/37,83.78% versus 13/35,37.14%,P 0.05).【Conclusions】 Both P53 protein and telomerase are involved in the process of gliomas etiology and progression,they might be new important markers of the malignant degree of brain glioma.
