摘要 目的从mRNA及蛋白水平检测DEK在正常宫颈、宫颈上皮内瘤变(CIN)及宫颈癌组织中的表达情况,探讨其在宫颈癌发病机制中的作用。方法采用RT-PCR及western-blot方法,检测DEK在35例宫颈癌组织、30例宫颈上皮内瘤变(CIN)及20例正常宫颈组织中的表达。结果宫颈癌组织中DEK mRNA和蛋白的阳性检出率分别为65.7%和71.4%,明显高于宫颈上皮内瘤变(分别为10.0%和16.7%)和正常宫颈组织(分别为5.0%和10.0%),差异具有显著性(P<0.05)。结论宫颈癌组织中存在着DEK原癌基因的激活,DEK基因激活和宫颈癌发生有关。 【Objective】 To detect the DEK expression in mRNA and protein levels in cervical cancer tissues,cervical intraepithelial neoplasia(CIN) and normal cervical tissues and to explore the pathogenesis of cervical cancer.【Methods】 DEK expression was detected in 35 cases of cervical cancer,30 cases of CIN and 20 cases of normal cervix by RT-PCR and Western blot.【Results】 The positive rates of DEK mRNA and protein in cervical cancer were 65.7% and 71.4%,respectively,which were significantly higher than those in CIN(10.0% and 16.7%) and normal tissue(5.0% and 10.0%)(P 0.05).【Conclusion】 DEK oncogene is activated in cervical cancer.DEK gene activation may be related to the occurrence of cervical cancer.
