摘要
目的研究亚硒酸钠对H2O2损伤PC12细胞的保护性作用。方法培养PC12细胞,加入亚硒酸钠,使其终浓度为0、0.5、1、5、8、10、20、50和150μg/mL,测定PC12细胞活力,选择不影响PC12细胞生长活性的亚硒酸钠浓度作为干预浓度,然后将细胞分为:正常对照组、损伤(+H2O2)组和亚硒酸钠保护组(+Na2SeO3+H2O2)组,采用甲基四唑蓝比色法(MTT)和流式细胞仪检测研究亚硒酸钠对H2O2氧化损伤PC12细胞的影响。结果①0.5μg/mL的亚硒酸钠对PC12细胞无抑制(P>0.05),随着浓度增大对PC12细胞的抑制率也增加(P<0.05)。②0.5μg/mL亚硒酸钠可以增强PC12细胞的抗H2O2氧化损伤的能力,提高细胞的生存率(P<0.05)。③经H2O2处理后,加入亚硒酸钠保护的细胞早期和晚期凋亡率较损伤组细胞有明显下降(P<0.05)。结论 0.5μg/mL的亚硒酸钠对PC12细胞具有抗H2O2氧化损伤的保护性作用。
【Objective】 To investigate the protective effect of Sodium Selenite against the oxidative damage of PC12 cell by H2O2.【Methods】 The PC12 cells were cultured,adding Sodium Selenite in them,accordance with the Sodium Selenite final concentration (0,0.5,1,5,8,10,20,50,150 μg/mL),divided into nine groups,determination viability of these groups .Choose the concentration which did not affect the activity of PC12 cell growth as the intervention concentration.Cells will be divided into three groups,the control group,damage group (+H2O2),Sodium Selenite protection group (+Na2SeO3+ H2O2).Using MTT method and flow cytometry studying the effect of Sodium Selenite on the oxidative damage to PC12 cells induced by hydrogen peroxide.【Results】 ① MTT method proved that the Sodium Selenite of 0.5 μg/mL has no inhibition effect to PC12 cell (P 0.05).With the increasing of Sodium Selenite's concentration,the inhibition effect will increase as well (P 0.05),② The Sodium Selenite of 0.5 μg/mL may strengthen the protective function of PC12 cells against the oxidative damage,improve the survival rate of cells (P 0.05).③ Both inchoate and late apoptosis rate of Sodium Selenite protection group are less than that of damage group.【Conclusion】 The results showed that the Sodium Selenite of 0.5 μg/mL has the protective function against the oxidative damage of PC12 cells which was made by H2O2.
