辽宁地区人群PPARγ 2基因Pro12Ala多态性与腹型肥胖的相关性研究

摘要 目的探讨过氧化物酶增殖体激活受体γ2基因(PPARγ 2)Pro12Ala多态性与腹型肥胖指数的关系。方法将研究对象健康对照者104人及2型糖尿病患者79人,按腰围/臀围比值(WHR)分为WHR正常组(121人)和WHR增高组(62人),应用聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)方法,检测所有研究对象、2型糖尿病患者和健康对照人群中WHR正常组和WHR增高组PPARγ 2基因Pro12Ala多态性,并对其基因型进行对照分析。结果①全部研究对象WHR增高组和WHR正常组基因型频率和等位基因频率比较差异无显著性(P值分别为0.46及0.701)。②2型糖尿病组中WHR增高组和WHR正常组基因型频率和等位基因频率比较差异无显著性(P值分别为0.524及0.531)。健康对照组中WHR增高组和WHR正常组基因型频率比较差异无显著性(P=0.055),但等位基因频率比较差异有显著性(P=0.022)。结论 PPARγ 2基因的Pro12Ala多态性与辽宁地区无T2DM家族史的正常人WHR增高有关,而与T2DM组WHR增高不相关。 【Objective】To determine the relationship between peroxisome proliferator-activated receptorγ2 gene (PPARγ2) Pro12Ala polymorphism and abdominal obesity index WHR.【Methods】 We included 183 individuals in this study: 79 type 2 diabetes mellitus (DM) patients and 104 non-diabetic control (NC) subjects.The study objects are divided into normal group and abnormal Group by the waist/hip ratio(WHR),Polymerase chain reaction (PCR)restrion fragment length polymorphism was used to determine the distribution of allele and genotype frequencies of Pro12Ala polymorphism in (PPARγ2) gene in 79 type 2 diabetes mellitus and 104 control subjects,50 normal WHR and 29 increased WHR subjects in DM group,71 normal WHR and 33 increased WHR subjects in NC group,differences in distribution between the groups were examined by χ2 test.【Results】Increased and normal group WHR genotype frequency,allele frequencies showed no statistical significance (P =0.46 and 0.701).The increased and normal group WHR genotype frequency,allele frequencies showed no statistical significance in DM group (P =0.524 and 0.531).The increased and normal group WHR genotype frequency showed no statistical significance in NC group (P =0.055),but allele frequency was significantly more (P =0.022).【Conclusion】 PPARγ2 gene Pro12Ala polymorphism may be related to the normal people who have abdominal obesity with increased WHR and without a family history of T2DM in the Liaoning region but not related to T2DM group who have abdominal obesity with increased WHR.
