高同型半胱氨酸致妊娠期高血压疾病作用机制的研究 被引量:3

摘要 目的探讨妊娠期高血压疾病患者同型半胱氨酸(Hcy)对细胞外超氧化物歧化酶(EC-SOD)表达的影响及其在PIH发生、发展中的作用机制。方法采用荧光偏振免疫分析法测定107例妊娠期高血压疾病患者和40例正常健康孕妇(健康对照组)空腹血浆Hcy的水平,根据测量结果将妊娠期高血压疾病患者根据Hcy水平分为妊娠期高血压疾病Hcy轻、中、重组,ELISA法等测定血清中GSH-px、SOD、MDA和ox-LDL的变化,实时RT-PCR、Western blot检测各组血浆中EC-SOD mRNA和蛋白表达。结果随着Hcy浓度的升高,妊娠期高血压疾病患者MDA和ox-LDL水平增加,GSH-px和SOD下降,EC-SOD的mRNA和蛋白表达下降。结论妊娠期高血压疾病患者Hcy水平的升高抑制EC-SOD的表达,导致氧化应激,是Hcy引发妊娠期高血压疾病形成的机制之一。 【Objective】 To study the mechanisms of homocysteine (Hcy) inducing extracellular superoxide dismutase mRNA and protein expression and oxidative stress in hypertensive disorders in pregnancy (PIH).【Methods】 The Hcy level of 107 pregnancy patients with hypertensive disorders and 40 health adults was measured by fluorescence polarization.PIH patients were divided into four groups by Hcy level: PIH Hcy normal group,PIH Hcy light range group,PIH Hcy midrange group and PIH Hcy serious group.The level of GSH-px,SOD,MDA and ox-LDL was detected by ELISA.EC-SOD mRNA and protein expression in blood plasma lymphomonocytes was measured by real time RT-PCR and Western blot respectively.【Results】 With the increase of Hcy level,the levels of MDA and ox-LDL in PIH patients were increased,the levels of GSH-px and SOD were decreased,the levels of EC-SOD mRNA and protein were lowered (P 0.05) in a dose-dependent manner.【Conclusion】 Homocysteine may play a crucial role in PIH by decreasing the expression of EC-SOD mRNA and protein and inducing oxidative stress.
