利用单抗免疫组化技术检测PCV-2 WH株在人工感染猪组织中的分布 被引量:5

Utilizing monoclonal antibody and IHC detect PCV-2 WH strains in different tissues of infected artificially swine

摘要 将猪圆环病毒2型(PCV-2)WH株通过颈部肌注和滴鼻感染26日龄PCV抗体阴性仔猪,于攻毒后28 d剖杀,采集相关的神经组织、消化器官及免疫器官进行免疫组化SABC法检测、病理学观察及PCR检测。免疫组化结果显示,在攻毒猪的大脑和小脑、胃、十二指肠、淋巴结、脾脏、扁桃体、肺、肾、肝存在阳性信号,心脏无阳性信号。阳性信号主要位于淋巴细胞和巨噬细胞的胞浆中,偶尔出现在细胞核内和上皮细胞中。免疫组化结果与病理学变化和PCR检测基本相符。结果表明,本试验建立的免疫组化SABC法,可用于定位PCV-2 WH株在体内不同组织器官中的分布情况。 Porcine cirovirus type 2(PCV-2) WH strain was injected into 26-day-old healthy pigs.After 28 days,the IHC,HE and PCR were used to detecte PCV-2 in neural and digestion organs taken from infected pigs at necropsy.IHC results showed the PCV-2 positive signals were detected at brain,cerebellum,stomach,duodenum,lymph nodes,spleen,tonsil,lung,kidney and liver organs,but negative in heart organ.The positive signals were mainly detected in cytoplasm of lymphocytes and macrophages and occasionally in the cell nucleus.The positive signals were also detected in epithelial cells.The result of three method have a great concordance.So the IHC SABC method is an alternative method which could be used for location of PCV-2 WH strain in different tissues.
