摘要
以墨兰的根状茎为外植体,研究培养基种类、NAA浓度和糖种类对墨兰根状茎增殖的影响.结果表明,当Hyponex1和Hyponex2单独使用时墨兰根状茎的生长及增殖效果不及MS培养基,但当Hyponex1和Hy-ponex2混合使用时明显好于MS培养基.BA和NAA的混合使用有利于根状茎的增殖生长,当Hyponex-1-2培养基中BA 2 mg/L的条件下加入0.2 mg/L NAA时,根状茎分化数多且鲜物重和干物重良好.糖种类对根状茎增殖生长影响较大,在Hyponex-1-2+BA 2 mg/L+NAA 0.2 mg/L培养基中加入30 g/L蔗糖时增殖效果最佳,其次是葡萄糖,果糖不利于根状茎增殖培养.
The rhizomes were used as explants to investigate the effect of medium types,NAA concentration and sugar types on rhizome proliferation and growth of Cymbidium sinense in vitro.The results showed that it is not suitable when Hyponex 1 and Hyponex 2 are individually used as culture medium for rhizome differentiation and growth,but combination of both media was optimum,better than MS medium.The medium supplemented with BA and NAA promoted rhizome proliferation,the maximum amount rhizome with favorable growth is found in the medium of Hyponex-12+BA 2 mg/L+NAA 0.2 mg/L.Sucrose was more suitable for rhizome culture than fructose and glucose.
