目的:探讨Survivin、VEGF-C在结肠癌发生、发展中的作用及相互关系。方法:免疫组化方法检测60例结肠癌组织和30例正常结肠组织中Survivin、VEGF-C的表达情况。结果:结肠癌组Survivin阳性表达率和VEGF-C阳性表达率均明显高于对照组(P<0.01)。结肠癌病理标本按浸润深度分组,浸润深度<5cm组Survivin和VEGF-C阳性表达率分别为60.6%、57.6%,浸润深度≥5cm组Survivin和VEGF-C阳性表达率分别为85.2%、81.5%,两组Survivin和VEGF-C阳性表达率均有明显差异(均P<0.05);按有无淋巴结转移分组,无淋巴结转移组Survivin和VEGF-C阳性表达率分别为60.0%、48.6%,有淋巴结转移组Survivin和VEGF-C阳性表达率分别为88.0%、96.0%,两组Survivin和VEGF-C阳性表达率也有明显差异(P<0.05和P<0.01)。VEGF-C和Survivin在结肠癌组织标本中的表达密切相关(r=0.40,P<0.05)。结论:Survivin和VEGF-C的表达异常是导致结肠组织细胞凋亡失控,异常增殖的主要原因。Survivin和VEGF-C的高水平表达在加速结肠癌向深处组织浸润与向周围淋巴结转移过程中具有协同作用。
Objective To study the possible role of Survivin and VEGF in the development of colonic carcinoma. Methods Expressions of Survivin and VEGF were examined with immunohistochemistry (IHC) in 60 specimens of colonic carcinoma and 30 non-malignant colonic tissue specimens. Results The expressions of Survivin and VEGF in the colonic carcinoma specimens were significantly higher than those in the non-malignant specimens. The positive rates of Survivin expression and VEGF expression in colonic carcinoma specimens with less penetration and infiltration (〈5cm, n=33) were 60.6% and 57.60% respectively, which were significantly lower than the corresponding expressions i.e. 85.2% and 81.5% in carcinoma specimens with deeper penetration and infiltration (5cm, n=27) (P〈0.05). The Survivin expressions and VEGF expressions in carcinoma specimens without lymphatic metastasis (n=35) were 60.0% and 48.6% respectively, which were also significantly lower than the corresponding ones (88% and 96%) in carcinoma tissues with metastasis (n=25, P〈0.05, P〈0.01). The expression rates of Survivin and VEGF in carcinoma specimens were mutually positively correlated (r=0.40, P〈0.05). Conclusion Abnormally high expressions of Survivin and VEGF in colonic tissue might be the main cause leading to excessive proliferation and defective apoptosis——together they favored development and progress of the malignant change.
