GnRHα超促排卵对围着床期小鼠子宫内膜LPAR3表达的影响

Effect of controlled ovarian hyperstimulation by GnRHα on the expression of LPAR3 in endometrium of mice during peri-implantation period

摘要 目的:研究GnRHα控制性超促排卵对围着床期小鼠子宫内膜溶血磷脂酸受体3(LPAR3)mRNA和蛋白表达的影响。方法:采用RT-PCR、Western blot方法检测自然受孕组(NC)和超促排卵组(COH)小鼠性交后第3~6天(3~6d.p.c)子宫内膜LPAR3mRNA和蛋白表达的表达。应用免疫化学发光法测定血清雌、孕激素浓度。结果:COH组3d.p.cLPAR3mRNA和蛋白的表达最强,NC组4d.p.c表达最强。其余相应各时间点两组间表达量差异无统计学意义;雌、孕激素在两组均随着妊娠天数增加而升高。相应同期各时间点COH组高于NC组(P〈0.05)。结论:COH可能通过改变与子宫内膜容受性相关的LPAR3的时序变化影响子宫内膜容受性,从而影响胚胎着床。 Objective:To investigate the effect of controlled ovarian hyperstimulation by GnRHα on the expressions of lysophosphatidic acid receptor 3(LPAR3)mRNA and protein in endometrium of mice during peri-implantation period.Methods:PT-PCR and Western blot were used to detect the expression levels of LPAR3 mRNA and protein in endometrium of mice at 3~6 days after sexual intercourse in natural conception group and controlled ovarian hyperstimulation group;the serum levels of estrogen and progesterone were detected by immunochemistry luminescence method.Results:The expression levels of LPAR3 mRNA and protein on the third day after sexual intercourse in controlled ovarian hyperstimulation group were the highest;the expression levels of LPAR3 mRNA and protein on the fourth day after sexual intercourse in natural conception group were the highest;there was no significant difference in the other days between the two groups;the serum levels of estrogen and progesterone in the two groups increased with the progress of pregnancy;at the same time points,the serum levels of estrogen and progesterone in controlled ovarian hyperstimulation group were significantly higher than those in natural conception group(P0.05).Conclusion:Controlled ovarian hyperstimulation may affect endometrial receptivity by changing LPAR3,which may affect implantation.
