摘要
目的:确定酶解法制备鹿茸胶原蛋白的最佳制备工艺。方法:用正交试验法考察胶原蛋白的提取工艺,用高效凝胶色谱法考察胶原蛋白的酶解条件,优选出鹿茸胶原蛋白提取的最佳工艺。结果:鹿茸胶原蛋白的制备工艺为:料液比(g:mL)1:3,水煮4h,提取5次,胰蛋白酶与底物(鲜鹿茸)的比是1:5 000,酶解时间为2h。结论:验证试验结果表明,鹿茸胶原蛋白收率为12.52%,蛋白含量为56.89%。本试验简便、稳定、可行。
Objective: To establish the optimal preparation technology of trypsin hydrolytic collagen from deer antler.Methods: Use orthogonal test to explore the extraction technology of collagen and explore the trypsin hydrolyze conditions by high performance liquid gel permeation chromatography,select the optimum preparation technology of trypsin hydrolytic collagen from deer antler.Results: The optimum preparation technology was obtained as follows: material-liquid rate(g:mL) was 1:3,boiled 4 hours,extracted five times,then hydrolyze 2 hours while the trypsin-substrate rate was 1:5000.Conclusion: The eventually deer antler collagen yield was 12.52%,protein content was 56.89%.It is confirmed that the extracting technology is simple,stable and feasible.
