摘要
目的:研究加味丹参饮对心肌缺血再灌注损伤(IRI)血瘀证兔内皮功能及内皮细胞超微结构影响,以探讨其对心肌IRI保护作用机制。方法:家兔60只,随机分为:空白组、血瘀证组、IRI组、预处理组、丹参组和加味丹参饮组,每组10只。建立血瘀证兔IRI模型,以加味丹参饮对该模型进行干预,采用放免法检测各组血浆ET-1和6-酮-PGF1a水平变化,透射电镜观察内皮细胞超微结构。结果:与空白组比较,IRI组ET-1水平明显升高,6-酮-PGF1a水平降低(P<0.01);与IRI组比较,加味丹参饮组可以降低ET-1水平,提高6-酮-PGF1a水平(P<0.01)。电镜观察显示加味丹参饮可以有效的改善心肌IRI时内皮的超微结构。结论:加味丹参饮能改善缺血再灌注损伤兔内皮功能,有效保护冠状动脉内皮细胞超微结构,从而对缺血再灌注损伤的心肌具保护作用。
Objective: To study the influence of jiawei danshen decoction (JDD)on endothelial function and ultrastructure of endothelial cell of myocardial ischemia reperfusion injury (IRI) in blood stasis syndrome (BSS) of rabbits, and to investigate protective mechanism of JDD on myocardial IRI. Method: Sixty rabbits were randomly divided into 6 groups with 10 rabbits each : control group ( A), BSS group ( B), IRI group ( C), Precondition group ( D), Danshen group ( E), and JDD group (F). A model of blood stasis syndrome of IRI was made and treated by JDD, and endothelin-1 ( ET-1 ) and 6-keto-prostaglandin F1α ( 6-Keto-PGF 1α ) in plasma were detected by a radio immune assay(RIA). Ultrastructure of endothelial cell was observed by an electron microscope. Result: Compared with the IRI group, JDD could significantly decrease the levels of ET-1, and increase the levels of 6-Keto-PGF 1α (P 〈 0. 01 ). JDD might improve ultrastructure of endothelial cell through observation under the electron microscope. Conclusion: JDD might improve endothelial function and ultrastructure of endothelial cell of myocardial ischemia repeffusion injury in blood stasis syndrome of rabbits. The results might display JDD' s prevention mechanism of myocardial IRI.
