摘要
目的观察橘荔散结丸有效部位对子宫肌瘤细胞增殖、凋亡的影响。方法采用胶原酶消化法培养子宫肌瘤细胞,采用免疫细胞化学方法鉴定子宫肌瘤细胞;将不同浓度的橘荔散结丸各提取部位组作用于子宫肌瘤细胞24 h后,用倒置显微镜观察细胞的形态变化,采用MTT法检测细胞的增殖活力,流式细胞术检测细胞的凋亡率。结果原代培养的子宫肌瘤细胞纯度接近100%;基质组对细胞活力无明显影响(P>0.05),而不同浓度的橘荔散结丸提取部位组能显著降低子宫肌瘤细胞的增殖活力(P<0.05),且呈浓度依赖关系(P<0.05);橘荔散结丸醇提物组则能提高其凋亡率(P<0.05)。结论橘荔散结丸的各提取部位组通过降低子宫肌瘤细胞的增殖能力,其醇提物组能诱导子宫肌瘤细胞凋亡而使子宫肌瘤自然缩小或消退,可能是橘荔散结丸治疗子宫肌瘤的主要机理之一。
OBJECTIVE To observe the impact on the proliferation and apoptosis of myoma cells exerted by the effective element in Juli Sanjie Pill.METHODS Collagenase digestive method was applied to the cultivation of hysteromyoma cells and chemical therapy of immune cells was to test them;Different components with diversified concentration of the pill acted on the hysteromyoma cells for 24 hours.In the experiment,inverted microscope was used to observe the morphological changes of the cells.MTT METHOD was adopted to test the growth vitality of the cells.In addition,FCM was used to test the apoptosis rate.RESULTS Cells' concentration was approximately 100% in primary cultivation group;There was no significant effect on cell vigor in matrix group(P〉0.05).Nevertheless,the extract of different concentration had evidently lowered the vitality of the cells relating to the concentration(P〈0.05),among which the alcohol extract group of the prescription enabled to enhance the apoptosis rate(P〈0.05).CONCLUSION Different experimental groups enable to decline the multiply capability of hysteromyoma cells.The alcohol extract group results in the natural shrinkage or removal of cells,which is probably one of the crucial mechanisms of Juli Sanjie Pill to cure hysteromyoma.
