金欣口服液含药血清及其有效单体白藜芦醇对RSV的RNA聚合酶转录活性的影响 被引量:19

摘要 目的研究金欣口服液含药血清及其有效单体白藜芦醇对呼吸道合胞病毒(RSV)RNA聚合酶转录活性的作用。方法通过体外分离病毒核糖核蛋白复合体(RNP)进行体外转录,并在转录过程中应用放射性核苷酸标记,同位素检测RSV的mRNA全长转录本,从而观察金欣口服液含药血清及其有效单体白藜芦醇对RSV的RNA依赖的RNA聚合酶活性的影响。结果正常细胞组与病毒感染细胞模型组放射性核苷酸辐射强度有显著差异(P<0.01),而白藜芦醇组转录掺入的放射性核苷酸明显低于病毒对照组(P<0.01),而各含药血清组差异不显著。结论金欣口服液有效单体白藜芦醇对RSV的RNA依赖的RNA聚合酶有较好的抑制作用,其可能是金欣口服液抗RSV的作用机制之一。 OBJECTIVE To study the effect of Jinxin Oral Liquid medicated serum and effective monomers Resveratrol on RNA polymerase transcription of respiratory syncytial virus(RSV).METHODS The ribonucleoprotein(RNP) complex of RSV was isolated and marked with radioactive nucleotides during the vitro transcription.The final incorporation of the radioactive nucleotides radiation intensity was detected to observe RNA-dependent RNA polymerase activity of RSV.RESULTS There was significantly statistical difference between normal cell group and RSV infected group as well as between RSV and resveratrol groups(P〈0.01).There was no significantly statistical difference among the medicated serum groups(P〉0.05).CONCLUSION Resveratrol makes an obvious inhibition on RSV's RNA polymerase,which may be one of the mechanisms of anti-RSV by Jinxin Oral Liquid.
