摘要
目的观察氟伐他汀对抗磷脂综合征(APS)患者血清条件下人脐静脉内皮细胞(HUVEC)组织因子(TF)表达的影响,并探讨其机制。方法50%APS患者血清作用于HUVEC24h,同时设立空白对照及正常血清(50%)对照组,不同浓度氟伐他汀(2.5、5.0和10.0μmol/L)干预。抗体夹心酶联免疫吸附试验(ELISA)检测各组细胞培养液中TF浓度,逆转录-聚合酶链反应(RT—PCR)检测各组细胞TFmRNA水平,Western blot免疫印迹法检测各组细胞核因子NF—KBp65含量。结果空白对照、正常血清组与APS患者血清组TF浓度、TFmRNA及NF—κBp65相对表达量分另U为(124.18±14.33)ng/L、(134.12±10.94)ng/L、(503.76±22.86)ng/L、(0.246±0.024)、(0.228±0.015)、(0.629±0.028)、(0.850±0.013)、(0.851±0.008)、(1.034±0.009)。空白对照与正常血清组比较差异无统计学意义(P〉0.05),APS患者血清组与上述两组比较差异有统计学意义(P〈0.05)。不同浓度氟伐他汀干预后TF浓度、TFmRNA及NF—κBp65相对表达量分别为(455.23±10.97)ng/L、(380.34±11.20)ng/L、(273.01±12.03)ng/L、(0.568±0.026)、(0.496±0.029)、(0.392±0.024)、(0.974±0.006)、(0.936±0.011)、(0.874±0.006),呈浓度依赖性下降(P〈0.05)。结论氟伐他汀能通过NF—KB抑制APS血清中抗磷脂抗体作用于内皮细胞后导致的组织因子增加。
Objective To investigate the effects of fluvastatin on tissue factor (TF) expression induced by serum of patients with antiphospholipid syndrome (APS) in human vascular endothelial cells (HUVEC). Methods HUVEC were treated with serum (50%) from APS patients, serum (50%) from normal persons, or culture medium ( blank control group), and interfered with different concentrations of fluvastatin (2. 5, 5.0 and 10. 0 μmol/L). Enzyme linked immunosorbent assay (ELISA) was used to examine the concentration of TF. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect TF mRNA. Western blotting was used to measure the content of nuclear factor-KB (NF-κB) p65. Results The concentration of TF and the relative expression of TF mRNA and NF-κB p65 in blank control group, normal serum group and APS serum group were (124. 18 ± 14. 33 ) , (134. 12± 10. 94), (503.76 _+22. 86) ng/L; (0. 246 ±0. 024), (0. 228±0. 015), (0. 629 ±0. 028) ; (0. 850 ±0. 013), (0. 851± 0. 008) , ( 1. 034 ± 0. 009) , respectively. There was no significant difference between blank control group and normal serum group ( P 〉 0.05 ) , but there was significant difference between APS serum group and other two groups (P 〈 0. 05 ). After treatment with different concentrations of fluvastatin (2. 5, 5.0 and 10. 0 μmol/L), the concentration of TF and the relative expression of TF mRNA and NF-KB p65 in blank control group, normal serum group and APS serum group were ( 455.23 ± 10. 97 ), ( 380. 34 ± 11.20 ), (273.01 ±12.03) ng/L; (0.568 ±0.026), (0.496 ±0.029), (0.392 ±0.024) ; (0.974±0.006), (0. 936 ± 0. 011 ) , (0. 874± 0. 006) , respectively, which were concentration-dependently reduced. Conclusion Fluvastatin displays a protective effect towards APS serum at the cellular level by decreasing TF expression in endothelial cells probably through the NF-κB pathway
