摘要
目的 了解丹参对大鼠血气改变和肺系数的影响以及丹参对肺泡巨噬细胞分泌肿瘤坏死因子( T N Fa) 、白介素- 1( I L- 1) 的调节作用。方法 静注油酸制造 A R D S大鼠模型为油酸组,静注生理盐水作对照组,静注油酸同时以丹参灌胃为治疗组。静注油酸及生理盐水6 小时后,测定左心 Pa O2 、 Pa C O2 及肺系数。分离培养各组大鼠肺泡巨噬细胞,采用 M T T 法测定 T N Fa、 I L- 1 。结果 油酸组 Pa O2 值明显低于对照组和治疗组,而 Pa C O2 值明显高于对照组和治疗组( P< 0001) 。治疗组血气明显好于油酸组( P< 005) 。油酸组肺系数(103 ±006) 高于治疗组(072 ±005) 和对照组(064 ±006) , P< 001 。 T N Fa 、 I L- 1 分别为油酸组(7206 ±564) % 、(9622 ±1161) U/ ml,对照组(775 ±105) % 、(2105 ±311) U/ ml,治疗组(3544 ±455) % 、(7066 ±823) U/ml。油酸组分泌 T N Fa 、 I L- 1 的水平显著高于对照组( P< 0001) ,治疗组亦显著高于?
Objective To explore the effect of Radix Salivae Miltiorrhizae on blood air and pulmonary weight index of rats suffuring from ARDS disease,and its regulating role on tumor nectosis factor(TNF a)and interleukins-l(IL-1) secretion in ARDS rats'alveole macrophages.Methods We make ARDS rats model by intraventing oil acid as the oil acid group.We take groups intravented with physiological saline as the control,and groups intravented with oil acid and simultaneously perfusated with Radix Salivae Miltiorrhizae into the stomach as treatment group.After intraventing oil acid and physiological saline for six hours.We measure the PaO 2 and PaCO 2 of the left heart and the pulmonary weight index.Then we separate and culture the alveolar macrophages of each group,and measure the quantities of TNF a and IL-1 by MTT method.Results The PaO 2 value of the oil acid group is more than the other two groups( P< 0 001).The bloods air of the treatment groups is better than the oil acid group( P< 0 05).The pulmonary weight indes of the three groups are oil acid group(1.03±0 06),the treatment group(0.72±0.05),the control group (0.64±0.06).There are significant differences among these groups( P< 0 01).The quantities of TNF a and IL-1 respectively are:the oil acid group(72.06±5 64)%、(96.22±11.61)U/ml,the control group(7.75±1.05)%、(21.05±3.11)U/ml,the treatment group(35.44±4.55)%、(70.66±8.23)U/ml.The secretion quantities of TNF a and IL-1 in oil acid group is far more than that in control group( P< 0 001).The treatment group also has higher level secretion than the control group,but its level lower than the oil acid group( P< 0 001).Conclusions Radix Salivae Miltiorrhizae can treat and protect ARDS rats with lung injury.It can also inhabit the excessive activation of alveloar macrophage in ARDS rats and reduce the secretion of TNF a and IL-1.The researches on the regulation of cellular factors secretion in alveolar macrophages are significant to recognize the pathogenesis of ARDS and guide clinical treatment.
