摘要
目的:将水性二相系统通过高速逆流色谱进行牛膝高分子物质中多糖成分的纯化研究。方法:以聚乙二醇与磷酸钾缓冲溶液组成水性二相系统,作为高速逆流色谱的固定相和流动相,对牛膝的高分子物质进行分离纯化。结果:125%聚乙二醇(PEG)1000与16%磷酸钾缓冲溶液(pH68)组成的水性二相系统,在XaxisCPC高速逆流色谱仪上,可以成功地分离出多糖部分和蛋白多糖部分。结论:借助高速逆流色谱,水性二相系统可以进行中药牛膝多糖的分离和纯化。
BJECTIVE: To purify polysaccharides from a highmolecular substance of Achyranthes bidentata root in an aqueous twophase system using highspeed countercurrent chromatography METHODS: An aqueous twophase system consisted of a polymer polyethylene glycol and an inorganic salt dibasic potassium phosphate in waterRegarded its lower phase and upper phase as the stationary phase and the mobile phase in countercurrent chromatography respectively RESULTS: The polysaccharides and proteoglycans were successfully separated by using an aqueous twophase system of 125% polyethylene glycol 1000 and 16% dibasic potassium phosphate buffer (pH 68) with the crossaxis coil planet centrifuge CONCLUSION: It is entirely possible to separate and purify the polysaccharides from the highmolecular weight substance of Abidentata root in an aqueous twophase system using highspeed countercurrent chromatography
