丁咯地尔片剂在家犬体内药代动力学和相对生物利用度

Investigation on pharmacokinetics and bioavailability of buflomedil tablets in healthy dogs

目的：比较丁咯地尔（甲氧吡丁苯）两种片剂在家犬体内药代动力学和相对生物利用度。方法：高效液相紫外检测法。结果：家犬口服后，其体内血药浓度时间曲线符合一室开放模型，其主要药代动力学参数分别为：ｃｍａｘ（１６．２４±４．９６）ｍｇ·Ｌ－１Ｌ和（１５．９１±５．９４）ｍｇ·Ｌ－１；ｔｍａｘ（０．８４±０．４５）ｈ和（０．８２±０．４９）ｈ；ＡＵＣ（９９．１３±２３．２２）ｍｇ·ｈ·Ｌ－１和（９７．９４±２５．６３）ｍｇ·ｈ·Ｌ－１。结论：国产甲氧吡丁苯片剂对进口甲氧吡丁苯片剂相对生物利用度为９８．５７％，可以认为国产片与进口片生物等效。

BJECTIVE: To compare the pharmacokinetics and bioavailability between domestic and imported Buflomedil tablets in dogs. METHOD: Buflomedil in plasma was determined by HPLCultraviollet method. RESULTS: It was found that the characteristics of the plasma Buflomedil concentrationtime curve fited to onecompartment model after a single oral dose of 80mgkg-1.The pharmacokinetic parameters of the two kinds of tablets were as follows:cmax16.244.96mgL-1;(15.915.94)mgL-1,tmax(0.840.45)h;(0.820.49)h,AUC(99.1323.22)mghL-1;(97.9425.63)mghL-1. CONCLUSION:BZThe relative bioava ailability of the domestic tablets was 98.57% compared with the imported ones.It showed that the two formn lations were bioequivalent.
