摘要
目的:建立用高效液相色谱法测定血浆中泮托拉唑浓度的方法,为进行泮托拉唑的体内药物代谢、生物利用度研究提供基础。方法:血浆用0.01mol·L-1磷酸氢二钠做pH调节剂,二氯甲烷做提取溶剂,硝西泮作内标。采用PlatinumC18(5u,250mm×4.6mm)柱,流动相:甲醇水醋酸盐缓冲液(40∶60∶5),流速1.0ml·min-1,检测波长286nm。结果:该法回收率平均在98.36%~104.27%,重现性高,RSD在4.693%~9.12%范围内(n=5),线性范围:20~4000μg·L-1,最低检测浓度15μg·L-1。结论:HPLC测定血浆中泮托拉唑的方法简便、准确、灵敏、专一,能满足血药浓度测定及药代动力学研究的需要。
BJECTIVE: To establish an assay to determine pantoprazole in plasma by means of highperformance liquid chromatography for the pharmacokinetic study of pantoprazole. METHOD: The platinum C18 column(250mm4.6mm,5u) was used as analytical column.The mobile phase consisted of methanolwateracetate buffer(pH4.3)40605.The flow rate was 1.00mimin-1.The blood samples were extracted by 0.01molL-1Na2HPO4 solution and dichloromethane,=286nm. RESULTS: The recovery of this method was 98.36%104.27% and the validation was RSD=4.69%9.12%(n=5). CONCLUSION: The method is simple, accurate and sensitive.It provides a suitable analytical method for its pharmacokinetic studies.
