Bcl-2、Bax、Cyt-C和Bid在原发性肝细胞癌中的表达及意义

Expression and Significance of Bcl-2,Bax,Cyt-C and Bid in Primary Hepatocelluar Carcinoma

目的观察Bc l-2、Bax、细胞色素C(Cyt-C)和B id在原发性肝细胞癌(PHC)组织中的表达并分析其与病理分期、肿瘤分化程度和淋巴结转移间的关系。方法应用免疫组织化学法检测Bax以及应用原位分子杂交方法检测Bc l-2、Cyt-C和B id在50例PHC组织及30例正常肝脏组织(对照组)中的表达。结果 PHC组Bc l-2、Bax和B id的阳性表达率较对照组均明显下降(P<0.05或0.01),但Cyt-C阳性表达率与对照组比较差异无统计学意义(P>0.05)。高及中分化PHC组Cyt-C阳性表达率均明显高于低分化组(P<0.05和P<0.01),但不同病理分级和有无淋巴结转移组间差异未见统计学意义(P>0.05)。PHC不同病理分级、分化程度和有无淋巴结转移组间Bc l-2、Bax和B id表达比较均未见统计学意义(P>0.05)。结论 PHC促凋亡基因B id表达下调,同时Cyt-C随着PHC分化程度的降低而表达下调。

Objective To observe the expression of Bcl - 2,Bax,Cytochrome C（ Cyt - C） and Bid in 50 cases of primary hepatocel- lular carcinoma （HCC group）and 30 cases of normal liver specimens （control group） ,and then to analyze their relationship with pathologi- cal grade,tumor differentiation and lymph node metastasis. Methods The expression of Bax was detected by immunohistochemistry, and the expressions of Bcl - 2, Cyt - C and Bid by hybridization in situ in 50 cases of primary HCC group and 30 cases of control group. Results The positive expression rate of Bcl - 2, Bax and Bid all decreased in HCC group as compared with control group （ P 〈 0.05 or 0.01 ）. The positive expression rate of Cyt- C didn＇t have significant difference between two groups （all P 〉 0.05 ）. The positive expres- sion of Cyt - C in high differentation and middle differentation sub-groups were higher than that in low differentation sub group,and there was significant difference （P 〈 0.01 and 〈 0.05）. However,the expression of Cyt- C had no relation with pathological grade and lymph node metastatis. The expression of Bcl -2, Bax and Bid didn＇t have significant difference in different pathological grades,tumor differenta- tion and lymph node metastasis groups （ all P 〉 O. 05 ）. Conclusion The expression of pro - apoptotic gene Bid decreased in PHC, and the expression of Cyt - C in PHC decreased with the decreasing of tumor differentiation extent.
