郁金香鳞片离体培养再生小鳞茎研究 被引量:7

摘要 以两个郁金香品种小天使和圣诞节的种球鳞片为外植体,研究不同接种方式、不同部位鳞片、不同基因型和激素浓度对郁金香鳞片离体培养再生小鳞茎的影响。结果表明,以鳞片凸面朝上进行培养,其成活率较高,达76.67%;以内层鳞片进行培养,其成活率达85.00%;小天使鳞片在添加1.0 mg/L 6-BA、0.5 mg/L NAA的MS培养基上的成活率和分化率均最高,而圣诞节在含2.0 mg/L 6-BA、0.5 mg/L NAA的MS培养基上的分化率较高;两个品种的小鳞茎在含3.0 mg/L 6-BA、0.2 mg/L NAA的MS培养基上增殖效果最好。 The bulb scales of two tulip cultivars,viz.,Little angel and Christmas were used to study the effects of different inoculation methods,different parts of bulb scale as explant,genotypes and hormone concentrations on in vitro regeneration of bulblet derived from different parts of bulb scale.The survival rate of bulb scales was recorded higher(76.67%) when the convexity of bulb scale was upward.The survival rate of inner bulb scale reached 85.00%.The bulb scale of variety Little angle had the highest survival rate and bulblet differentiation rate,when cultured on MS medium supplemented with 6-BA(1.0 mg/L) and NAA(0.5 mg/L),while variety Christmas had higher bulblet differentiation rate on MS medium supplemented with 6-BA(2.0 mg/L) and NAA(0.5 mg/L).The induced bulblet of two varieties showed best proliferation on MS medium supplemented with 6-BA(3.0 mg/L) and NAA(0.2 mg/L).
