澳大利亚香蕉品种Williams球茎再生体系研究 被引量:1

Development of regeneration system for corms of banana Williams introduced from Australia

摘要 以澳大利亚香蕉品种Williams吸芽球茎种植获得的球茎为外植体,探讨不同激素组合对球茎萌芽诱导、增殖分化、生根以及不同基质对生根苗移栽的影响。结果表明,球茎在初代培养基MS+6-BA 3.5 mg/L+NAA 0.1 mg/L+蔗糖30.0 g/L+琼脂3.0 g/L中培养18 d后即可萌芽生长,生长速度快,长势强;在培养基中添加不同浓度MT的增殖效果优于6-BA,且芽体分株清晰,高度适中,无愈伤组织,芽体整齐一致,继代芽容易切割,增殖系数高,其中0.5 mg/L MT有利于促进芽体长根。在MS培养基中添加IBA、NAA均可有效诱导芽苗生根,根系生长较旺盛。在不同移栽基质泥炭土、沙土混合、田园土中,生根苗移栽成活率均达95.0%以上。大田种植结果表明,4月种植的Williams组培苗在8月假茎高为195.00 cm,累计叶片数为24.67张,最大叶长和叶宽分别为201.33和80.33 cm,其农艺性状表现较好。 The corms of banana Williams plantlets derived from corm of axillary buds were used as explant to study the effects of different hormone combinations on germination,proliferation and differentiation,rooting,and transplantation of rooting plantlets.The corms showed good germination and strong growth vigor,when cultured in MS medium supplemented with 6-BA(3.5 mg/L),NAA(0.1 mg/L),sugar(30.0 g/L) and agar(3.0 g/L) for 18 days.The germinating buds showed better proliferation when cultured in MS medium supplemented with MT compared to 6-BA.The MS medium with 0.5 mg/L MT showed better effects on promoting the rooting of germinating buds.The MS medium supplemented with IBA and NAA could induce the rooting of bud.The rooting plantlets were found to grow well in different transplanting media,viz.,peat soil,mixture of sand and soil,and field soil.The survival rate of plantlets was recorded more than over 95.0%.After transplanted in April,the cauloid height of plantlet in August reached 195.0 cm,and the number of leaves,the max leaf length and width was 24.67,201.33 and 80.33 cm,respectively.
