不同偶联方式的水溶性多糖高分子前药的应用

Application of water-soluble polysaccharide polymer prodrug conjugated in different ways

水溶性多糖同药物以不同的偶联方式制得的高分子前药可以减轻药物的毒副反应,提高药物的疗效和稳定性,可以靶向给药以及实现对药物的缓控释放.本文从偶联方式的角度综述了其应用,偶联方式主要包括直接偶联(酯键,酰胺键连接),连接臂间接偶联(丁二酸,戊二酸,Click化学生成的三氮唑等),咪唑中间体偶联等,最后对不同偶联方式的水溶性多糖高分子前药的应用前景做了展望.

OBJECTIVE Water-soluble polysaccharide polymer prodrugs conjugated in different ways can reduce side effects and improve bioavailability and stability of drug,to targeted delivery and control time to release drug and distribution.This article will discuss its application from different ways of conjugated,including direct coupling（ester,amide）,indirect coupling with introduction of spacers（succinate,glutaric acid,triazole from Click chemistry）,Imidazole intermediate coupling and so on.At last,future trends of the application of water-soluble polysaccharide conjugated in different ways polymer prodrug is prospected.
