藏药“藏茵陈”的位点特异性PCR鉴别(英文)

Allele-Specific Primers for Diagnostic PCR Authentication of Swertia mussotii the Traditional Tibetan Medicinal Plant

对与藏药"藏茵陈"药材质量、真伪密切相关的药材来源进行鉴别.首先考察、鉴定市售"藏茵陈"药材伪品原植物种,通过对这些植物进行分子序列分析,来比较正品原植物与伪品植物的分子序列碱基位点和分子片段的区别,从而筛选出可靠的分子标记来鉴别"藏茵陈"原植物川西獐牙菜Swertia mussotiiFranch.的真伪.根据"藏茵陈"原植物特异性的分子片段,设计专用于鉴别川西獐牙菜的PCR引物,用该引物在不同条件下扩增正品及伪品"藏茵陈"模板DNA,建立只有正品川西獐牙菜具有阳性扩增的PCR鉴别条件.从而建立一种简便、适用的"藏茵陈"川西獐牙菜的DNA分子鉴别方法.

Compared with drug quality and those similar to ＂Zangyinchen＂,identification research has been done to appraise the fake species plant sold in the market by which molecular sequence analysis has been finished to compare the differences of molecular sequence base pair and molecular fragment between genuine and sham ＂Zangyinchen＂ so as to screen out the original plant Swertia mussotii of ＂Zangyinchen＂ by the reliable molecular tag.According to molecular fragment specificity of original plant of ＂Zangyinchen＂,a special primer to identify PCR of Swertia mussotii has been designed and under different conditions,using the diagnostic primers with the total DNA of the original plants as a template to set up positive cloning PRC identifying condition only possessed by genuine Swertia mussotii to get simple and practical DNA identifying method.
