摘要 目的:研究不同浓度IL-1β干预下KGF在人牙龈成纤维细胞(HGFs)炎症损伤过程中基因表达水平的变化。方法:取原代培养至5~7代的HGFs,以IL-1β梯度浓度干预,采用RT-PCR方法检测KGF在HGFs中的表达情况。结果:在正常的HGFs中KGF呈弱阳性表达,随着IL-1β的浓度由0.1 ng/ml增加到10.0 ng/ml,KGF表达逐渐增高。结论:KGF在IL-1β引发的HGFs炎症过程中可能发挥重要的作用,为了解牙周病的发病机制提供新的资料。 Objective: To investigate the expression of keratinocyte growth factor(KGF) during the damaging process of human gingival fibroblasts(HGFs) under the intervention of IL-1β with different concentrations.Methods: From 5th to 7th generation of the HGFs culture was treated with IL-1β of serial concentrations.The expression of KGF in HGFs was examined by RT-PCR method.Results: KGF was weak positive expression in normal HGFs.With the increasing concertraion from 0.1 ng/ml to 10.0 ng/ml of IL-1β,the expression of KGF gradually increased.Conclusion: KGF plays an important role during inflammatory process of HGFs caused by IL-1β.Then for understanding the pathogenesis of periodontal disease provide new informations.
