摘要
分别用以DEAE.Sephadex A-50离子交换层析和Sepharose 4B-肝素钠亲和层析纯化猪凝血酶,对2种方法纯化结果进行比较。结果表明,离子交换层析纯化的凝血酶比活为1132IU/mg,纯化11.1倍,收率为45.3%;亲和层析的酶比活为1789IU/mg,纯化17.5倍,收率为58.3%;亲和层析纯化效果明显优于DEAE—SephadexA-50离子交换层析。
Porcine thrombin was purified by DEAE-Sephadex A 50 ion-exchange chromatography and Sepharose 4B-hep- arin affinity chromatography and the purification results were compared. The thrombin activity, purification fold and yield of purified thrombin by DEAE-Sephadex A 50 ion-exchange chromatography are 1132 IU/mg, 11.1 folds and 45.3% and those by Sepharose 4B-heparin affinity chromatography are 1789 IU/mg, 17.5 folds and 58.3%. The method of Sepharose 4B-heparin affinity chromatography is superior to that of DEAE-Sephadex A 50 ion-exchange chromatography.
