摘要
本文对利用花粉管通道技术将耐盐碱罗布麻(Apocynum venetum)DNA导入陆地棉(Gossypi-um hirsutum L)品种鲁棉6号从后代中筛选出的耐盐变异体山农011进行RAPD分析,140个引物中有2个引物扩增出多态性产物,表明供体罗布麻DNA的导入引起受体基因组的变异;对耐盐变异体的抗盐生理研究表明:盐胁迫下,山农011具有较高的渗透调节能力,其渗透调节能力的提高是以细胞内渗透调节物质(如脯氨酸)的积累为基础.
The salt-tolerance variation of cotton-Shannong 011 obstained from the offsprings of introducing bluish dogbane (Apocynum venetum) DNA into the Lumian No. 6(Gossypium hirsu-tum) by pollen tube pathway, was used for RAPD analysis. 140 primers used for detecting the polymorphism. Only two primers (OPX-18、OPY-18) gave polymorphic amplification products. These results showed the introduction of dogbane DNA might cause the variation of recipient genome. Also, physiologic studies indecated that the increase in salt resistance of the variation was for a result of the decrease of osmotic potential and the increase of osmotic adjustment activity, and this result was based on enhancement of organic osmotors.
