摘要
目的探讨氧化应激在三氯乙烯(trichloroethylene,TCE)致敏豚鼠肝损伤中的作用。方法选用体重250-300 g的白色雌性豚鼠,随机分成空白对照组,溶剂(橄榄油)对照组,TCE处理组。根据豚鼠最大值实验(guinea pig maximization test,GPMT)方法处理豚鼠。按照《化学品毒性鉴定技术规范》的评分标准对动物的皮肤反应进行评分,评分≥1的判为致敏。在终末激发后24 h和72 h取材,对肝组织进行病理学检查,并分别测定血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和白蛋白(ALB)和肝脏中的丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力。结果TCE处理组致敏率为65.38%。TCE致敏72 h组中见到较多肝细胞水肿,并有胞核破裂消失。TCE致敏72 h组ALT和AST水平比TCE致敏24 h组和TCE未致敏72 h组升高,差异有统计学意义(P〈0.05);TCE致敏72 h组ALB水平比溶剂对照组和TCE未致敏72 h组降低,差异有统计学意义(P〈0.05)。TCE致敏24 h和72 h组肝MDA含量与溶剂对照组相比明显升高,且差异有统计学意义(P〈0.05);TCE致敏24 h组肝SOD活力与溶剂对照组相比较明显降低,差异有统计学意义(P〈0.05),而TCE致敏72 h组肝SOD活力与溶剂对照组相比较明显升高,差异有统计学意义(P〈0.05)。结论TCE可引起致敏豚鼠肝损伤,氧化应激在肝损伤中发挥一定的作用。
Objective To Investigate the effect of oxidative stress on liver damage in trichloroethylene(TCE)sensitized guinea pigs.Methods White female guinea pigs(250—300 g)were randomly divided into blank control group,solvent(olive oil)control group and TCE treatment group.Guinea pigs were treated with Guinea Pig Maximization Test(GPMT).According to "Chemical Toxicity Appraisal Technology Standard",the animal was considered as sensitized if the skin reaction score was greater than or equal to 1.The liver samples were taken at 24 h and 72 h after last stimulation respectively,then performed pathological examinations and detected AST,ALT activities and ALB levels in serum,and MDA level,SOD activity in liver with kits.Results The sensitization rate was 65.38% in TCE treatment group.Rats in TCE sensitized 72 h group could be seen more cellular edema in liver,and some nucleus rupture or effacement.The levels of ALT and AST were increased significantly(P0.05)compared with TCE sensitized 24 h group and 72 h TCE un-sensitized group;while the level of ALB was decreased significantly(P0.05)compared with TCE solvent control group and TCE un-sensitized 72 h group.The MDA levels in liver of both 24 h and 72 h TCE sensitized groups were significantly higher(P0.05)compared with solvent control group;the SOD activity of the liver in TCE sensitized 24 h group was decreased compared with the control group,while the SOD activity of liver in TCE sensitized 72 h group was significantly higher(P0.05).Conclusion TCE can cause liver injury in sensitized guinea pigs,the oxidative stress plays some role in liver injury by TCE.
