摘要
目的:观察秦香颗粒(秦香)对HRV感染乳鼠小肠黏膜淋巴细胞的影响,探讨秦香颗粒治疗人轮状病毒(HRV)感染性腹泻的可能作用机制。方法:将乳鼠随机分成7组,正常组常规饲养,其余6组造成HRV感染模型,并分别ig生理盐水、秦香颗粒高、中、低剂量(1.44,0.72,0.36 g·kg-1)、病毒唑(0.11 g·kg-1·d-1)及肠康片(0.52 g·kg-1·d-1),连续给药5 d。采用金标法检测粪便HRV抗原,免疫组化法观察小肠黏膜淋巴细胞的变化。结果:与正常组比较,各治疗组及模型组小肠黏膜淋巴细胞CD4+减少(P<0.05);与模型组比较,各给药组小肠黏膜淋巴细胞CD4+的数量不同程度上升,其中秦香颗粒高、中剂量组及病毒唑组具有统计学意义(P<0.05);与正常对照组比较,各给药组及模型组小肠黏膜淋巴细胞CD8+数量不同程度增加,与模型组比较,各组小肠黏膜淋巴细胞CD8+的数量不同程度减少,差异均具有统计学意义(P<0.05)。结论:秦香颗粒可以上调HRV感染乳鼠小肠黏膜CD4+、下调CD8+,有利于增强淋巴细胞对肠黏膜的保护作用,减轻肠黏膜病变,对HRV感染性腹泻有一定的治疗作用。
Objective:To observe Qinxiang granules(Qinxiang Zhixie tablets) influence on small intestinal mucosa lymphocytes in suckling mice with HRV infection.Method:The suckling mice were randomly divided into 7 groups,the normal control group,the other six groups as the model of HRV infection.The normal control group was treated with NS,and the other experimented groups differently treated with Qinxiang granules(High dose/medium dose/low dose,1.44/0.72/0.36 g·kg-1),ribavirin(0.11 g·kg-1),Changkang tablets(0.52 g·kg-1) for 5 days.The changes in small intestinal was observed.Result:Compared with the normal control group,in each treatment group and model group reduction of small intestinal CD4+lymphocytes was statistically significant(P0.05).Compared with model group,in the treatment groups small intestinal CD4+lymphocytes were increased in high,medium dose groups and ribavirin group the increase was significant(P0.05);comparied with normal control group,in each treatment group and model group,the number of small intestinal CD8+lymphocytes was increased to various degree.Compared with model group,in the treatment groups small intestinal CD8+lymphocytes werereduced,and the differences were significant(P0.05).Conclusion: Qinxiang granules can increase small intestinal CD4+,lower CD8+in suckling mice.The Qinxiang granules is conducive for enhancing the protection of the intestinal mucosa with lymphocytes,and reducing mucosal lesions.
