摘要
本文采用正交试验对法夫酵母高密度培养条件进行了研究,得到其优化培养基组成为:葡萄糖35g/L、酵母膏9g/L、KNO30.6g/L、(NH4)2SO4 0.5g/L、KH2PO4 1.5g/L、MgSO4 0.2g/L,最适摇瓶装液系数为10%。在10L罐上的实验结果表明:采用优化后的培养基在48h细胞总数就可以达到1.5×109个/mL,与初始培养基相比细胞总数提高了88.7%,到达最高菌体浓度提前了12h。
The high density fermentation of Phaffia rhodozyma was studied by o rthogonal test. The results showed that the optimal fermentation medium contained: sucrose 35g/L, yeast extract 9g/L, KNO3 0.6g/L, (NH4)2SO4 0.5g/L, KH2PO4 1.5g/L and MgSO4 0.2g/L. The optimal medium volume in flask fermentation was 10%(v/v) of the flask. The results of the fermentation in 10L fermentor showed that the cell density reached 1.0×10^9cells/rnl after being cultured in the optimal medium for 48h, which was increased by 88.7% than that of the original medium.
