UFLC法测定母丁香中丁香酚和2-羟基-4,6-二甲氧基-5-甲基苯乙酮的含量 被引量:6

UFLC determination of eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone in Eugenia caryophyllata Thunb.

摘要 目的:建立母丁香中丁香酚和2-羟基-4,6-二甲氧基-5-甲基苯乙酮的超快速液相色谱(UFLC)含量测定方法,完善母丁香药材的质量标准。方法:采用Shim-pack XR-ODSⅡ(3.0 mm×75 mm,2.2μm),柱温50℃,流动相为甲醇-0.5%磷酸水溶液(60∶40),流速0.8 mL.min-1,检测波长280 nm。结果:丁香酚和2-羟基-4,6-二甲氧基-5-甲基苯乙酮进样量分别在9.98~79.84μg和17.08~136.64μg范围内与峰面积呈良好的线性关系(r=0.9999),平均加样回收率(n=5)分别为99.57%和98.68%。结论:母丁香药材的UFLC含量测定法快速、准确、简便、可靠,是一种值得推广的中药质量评价方法。 Objective:To a ultra-fast liquid chromatography(UFLC) method for the determination of eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone in Eugenia caryophyllata.Methods:Shim-pack XR-ODS Ⅱ(3.0 mm×75 mm,2.2 μm) column was adopted with methanol-0.5% phosphoric acid water solution(60∶40) as the mobile phase at a flow rate of 0.8 mL·min-1;The detection wavelength was 280 nm and column temperature was 50 ℃.Results:Both eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone showed a good linear relationship with peak area within the range of 9.98-79.84 μg(r=0.9999) and 17.08-136.64 μg(r=0.9999),respectively;The average recoveries(n=5) were 99.57% and 98.68%.Conclusion:The UFLC method that is used to quantify eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone is simple,rapid,accurate,and good reproducibility,so it can be used to evaluate the quality of Eugenia caryophyllata;UFLC method can save analysis time and solvent consumption,thus it can be worthy of promotion to quality the traditional Chinese medicine.
