摘要
目的:建立再造丸的定性定量方法。方法:采用TLC法对方中的人工牛黄、大黄、当归和川芎、麻黄进行定性鉴别;采用TLC法对乌头碱的限量进行检查;采用HPLC法测定再造丸中天麻的活性成分天麻素的含量:使用岛津Shim-pack VP-ODS(150 mm×4.6 mm,5μm)色谱柱,以甲醇-磷酸盐溶液(含磷酸氢二钠和磷酸二氢钾各0.1 mol.L-1)-水(0.5∶3∶96.5)为流动相,流速1.0 mL.min-1,检测波长221 nm,柱温30℃。结果:TLC色谱中可检出人工牛黄、大黄、当归和川芎、麻黄;天麻素进样量在0.0428~1.284μg范围内,线性关系良好,平均回收率为95.5%(RSD=1.2%,n=6)。结论:该法准确,重复性好,可用于再造丸质量控制。
Objective:To establish the methods for qualitative and quantitative of Zaizao pills.Methods:Calculus Bovis,Radix et Rhizoma Rhei,Radix Angelicae Sinensis and Rhizoma Chuanxiong,Herba Ephedrae were identified by TLC.Limit test for aconitine was examined by TLC.The content of gastrodin was determined by HPLC.HPLC was performed on a Shimadzu Shim-pack VP-ODS(150 mm×4.6 mm,5 μm) column with methanol-phosphate solution(containing 0.1mol·L-1 disodium hydrogen phosphate and 0.1 mol·L-1 potassium dihydrogen phosphate)-water(0.5∶3∶96.5) at the flow rate of 1.0 mL·min-1,detected at 221 nm.The column temperature was at 30 ℃.Results:Calculus Bovis,Radix Angelicae Sinensis and Rhizoma Chuanxiong,Radix et Rhizoma Rhei,Herba Ephedrae could be identified by TLC.The gastrodin showed a good linear relationship at the range of 0.0428-1.284 μg,the average recovery(n=6) was 95.5% with RSD of 1.2%.Conclusion:The method is accurate with a good reproducibility,and can be used for the quality control of Zaizao pills.
