摘要
目的:探讨灯盏乙素对过氧化氢(H2O2)损伤的血管内皮细胞的保护作用。方法:体外培养血管内皮细胞,将细胞分为6组,即正常对照组(control),氧化损伤组(H2O2组),氧化损伤加入维生素E对照组(VitE+H2O2),氧化损伤加入灯盏乙素组(scutellarin1+H2O2,scutellarin2+H2O2,scutellarin3+H2O2)。用VitE及不同浓度灯盏乙素预先培养血管内皮细胞24 h,然后加入1 mmol.L-1 H2O2继续培养24 h,MTT法检测细胞存活率;检测SOD,GSH-PX,CAT活力。结果:不同浓度灯盏乙素呈能够降低H2O2对抗氧化酶SOD,GSH-PX,CAT活力的影响,各指标差异有显著性(P<0.01)。结论:灯盏乙素可保护过氧化氢诱导的血管内皮细胞的损伤,其作用可能与抗氧化、增强抗氧化酶SOD、GSH-PX、CAT的活力有关。
Objective: To explore the protective effects of scutellarin on vascular endothelial cell injury induced by oxidative hydrogen.Methods: Preincubate with different concentrate of scutellarin and VE,1 mmol·L-1 Peroxide hydrogen(H2O2) was used to induce injury of human umbilical vein endothelial cells(HUVEC).The cell survival rate was evaluated with MTT assay to explore the protective effect of scutellarin on injury of vascular endothelial cells.The activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX) and catalase(CAT) was detected in H2O2-induced HUVEC injury.Results: HUVEC was incubated with 1 mmol·L-1 H2O2 for 4 h,the cell viability decreased significantly(P﹤0.05 vs normal control group);scutellarin can promote the cell viability of HUVEC induced by H2O2(P﹤0.05 vs H2O2-injury group);scutellarin could concentration-dependently protect the activity of SOD and GSH-PX(P﹤0.05 vs H2O2 injury group).Conclusion: Scutellarin showed protective effects on vascular endothelial cells induced by oxidative stress and the probable mechanisms may be closely associated with its promoting the activity of antioxidases.
