摘要
目的:探讨肿瘤坏死因子( T N Fα) P G E2 ,在急性肝损伤中的作用和肝苏冲剂的保肝作用机制。方法:利用 C C I4诱导小鼠急性肝损伤动物模型,观察了模型小鼠血清及肝组织匀浆中丙氨酸转氨酶( A L T) 、 T N Fα、 P G E2 及肝脏病理变化。结果:模型组肝组织明显受损, A L T 与 T N Fα显著增高。模型+ 肝苏冲剂组肝组织受损较轻, A T L、 T N Fα水平明显降低, P G E2 水平增高。结论: T N Fα在急性肝损伤中起重要作用。肝苏冲剂可通过提高 P G E2 水平,抑制 T N Fα产生,发挥保肝作用。
Objective: To study the effect of tumor necrosis factor-α(TNFα)、prostaglandin E2(PGE2) and the protective effect of GANSUCHONGJI (GSCJ)in acute liver injury .Methods:The mouse model of experimental hepatic injury was made by the inducement of CCl 4. We detected the changes of alanine aminotransferase(ALT)、TNFα、PGE2 in liver tissue,as well as the pathological changes of the liver.Results:The levels of ALT、TNFα in the model group were significantly higher than that of the control group(P<0.01).The levels of ALT、TNFα,however,were significantly decreased in the model group with GSCJ protection.Meanwhite,the level of PGE2 in GSCJ group was significantly higher than that of the model group. Conclusion: TNFα plays an important role in liver injury.One of the mechanisms of hepatic protection of GSCJ lies in the enhancement of PGE2 and the inhibition of TNFα production.
