摘要
目的探讨乳腺癌组织中Notch1受体的表达水平,为乳腺癌的防治提供新的策略。方法进行荧光定量PCR(FQ-PCR)和免疫组化技术对60例乳腺癌组织和25例正常乳腺组织中的Notch1受体蛋白及基因表达情况的检测分析。结果 (1)FQ-PCR技术检测显示乳腺癌患者Notch1基因表达阳性,DNA拷贝数显著高于对照组,差异有显著性(P<0.05);侧腋窝淋巴结转移组Notch1 DNA拷贝数稍高于非转移组,但差异无显著性(P>0.05)。(2)免疫组织化学染色的结果表明Notch1分子在乳腺癌组织中呈强表达,正常乳腺组织中不表达或弱表达,差异有显著性(P<0.05)。结论 Notch1信号在乳腺上皮干细胞恶性转化中表达上调,Notch1基因可能是一种癌基因,对乳腺癌干细胞的分化起调节作用。
Objective To investigate the role of Notch1 receptor in human breast carcinoma tissue in the prevention and treatment of liver cancer. Methods Sixty cases of invasive ductal carcinoma were collected in this study and RT-PCR and immunohistochemistry methods were used to detect the expression level of Notchl receptor in human breast carcinoma and normal breast tissue, followed by statistical analysis. Results The gene expression of Notchl was significantly up-regulated in human breast carcinoma (P〈0.05). The result of immunohistochemical staining was similar to that of the RT-PCR,and the DNA copy number of Notchl in the lateral axillary lymph node metastasis was slightly higher than that of non-metastasis,with no significant difference(P〉0.05). Conclusion During the malignant transformation of mammary stem cells,the Notchl expression is up-regulated. Notchl gene may be an oncogene candidate, which may regulate the differentiation of mammary cancer stem cells.
